Hi there,
First travel from Shinjuku to Shinagawa or Tokyo stations to get on the Shinkansen bullet trains. Then there is one transfer at Shin-Osaka from the Hikari to the Sakura Shinkansen.
The full trip is covered by the JR Pass.
