JR trains loop in Tokyo
Shinagawa Station to Osaka
Osaka to Villa Fontaine Hotel
Local train with in Osaka

The JR Pass covers travel between Tokyo <-> Osaka, local JR lines in Tokyo and Osaka as well as a lot more.
However, you may still need additional local transport depending on what you want to see/do.
