reserve seat?

Seat reservations are completely free with the JR-Pass and can be made at any ticket office or manned ticket counter. Simply show your JR-Pass and tell the staff where you would like to go.
You can make your first seat reservations when you exchange your JR-Pass.

Yes the journey between Kansai Airport and Osaka is fully covered and you can use the Haruka limited express too!

So if I understand well, we can reserve, only the date of the exchange and only in Japan, no from France 2 month before our travel. Because i'm afraid not to have seat available on "spring holidays starts on April 27 and will probably end on May 6".

Yes in general you can only make reservations once you are in Japan. However, I don't think you will have to worry much about reservations.
Japan school holidays are a month before from around the end of march until the 1st week of April.
Even during this period though if you make reservations a day before departure you should have no problem.
