I will be going to osaka on the 5/2 arriving at night.

If you don't return from Tokyo it is probably better to just buy separate tickets. The travel time between Kyoto and Tokyo is about 160 Minutes by the Hikari Shinkansen.
I am not entirely sure what you mean by ''Do you think that is enough?'' but if you mean to have a good time in Japan, then Yes! Consider adding a day to Kyoto as it is a nice switch from the metropolitan cities like Tokyo and Osaka.
