We will be arriving Kansai on April01 and will leave Narita on April 09.
01 & 02 - Osaka
03 - Osaka - Kyoto - Osaka
04 - Osaka - Tokyo
05 to 09 - Tokyo
1) Where can we purchase the Suica + one way N'EX since we will not be arriving at Narita?
2) Can we use the 3day JR East Pass Special (10,000yen) from Osaka to Tokyo Shinkansen?

I think the Suica + One Way NEX is only valid for purchase at Narita. But you could try purchaseing it at the ticket office at Tokyo station on arrival
2) Can we use the 3day JR East Pass Special (10,000yen) from Osaka to Tokyo Shinkansen?
I'm afraid not. The JR East Pass special is only valid for trains in the JR-East/Tohoku area. As the Tokyo - Osaka bullet train passes through the JR-East, JR-Central and JR-West zones, I'm afraid the East Pass Special will not be valid.
