Tickets in the Kansai Area are not very expansive in general.
I would recommend that you buy single tickets as you go for the above routes.

Since you are arriving at Kansai Airport, a Haruka + Icoca package may be of interest. It offers a smooth ride into Osaka and comes with a rechargeable IC-Card that you can use for your further travels in the Area.

Since we are traveling during the cherry blossom season, will the train ticket lines be very long if we have to buy single tickets each time?

Yes trains may be a little more busy that normal, if you buy an IC card you can just swipe it over the ticket gate and you won't have to queue up for the ticket machines. In Kansai the most widely accepted IC card is Icoca. I don't think you will have to wait very long though, if you don't use one.
